How is it that I can deny a user from posting in a specific thread is the user doesnt have X amount of reputation/post?

You could do it with usergroup permissions and promotions, except that permissions only apply to forums and not to individual threads. You could probably write a fairly simple plugin that is called when a user posts and checks the threadid and the user's post count before allowing the post.
