Spam-o-matic filters based on user registrations. Anything that has already happened is a done deal. If you find users who have spammed in posts or just by the content of their sig file, you can use the spam-o-matic menu in the user profile to ban them and submit their info to the stopforumspam database. As far as I know, weeding out existing spam users is a manual process.
