It really does depend on how many clients you have on your site and what they are doing. In order of priority and on a very general basis: RAM, CPU, Hard Disk.

Based upon 500 users online, caching and clustering should yield high benefits. In addition, a php accelerator will lower your load. We have a client with 500-1000 users online at any time and they use Memcache, eAccelerator, clustering and SCSI RAID and their site is quite snappy no matter how many users are on.
