Just delete the one(s) you are replacing. Definitions do not overwrite. The Master is for people starting new or starting over.

Since upgrading I've been experiencing problems w/ Photobucket videos. It appears as though it's using the embedd code so there are 2 squares for the video. The top square is empty and the bottom is the video.

Sounds to me like you have two Photobucket definitions.

Looks like gametrailers def no longer works fine. Now most urls there have game name in url like:
http://www.gametrailers.com/video/vs...smash-up/56351
http://www.gametrailers.com/video/re...-turismo/56347
Same foe user movies:
http://www.gametrailers.com/user-mov...an-gt-2/164801
and current definitions don’t handle it well :( looks like it need to be updated. Since it only support urls like:
http://www.gametrailers.com/player/56351.html
