Where can I change the layout of Vbulletin Options

You mean the layout of your AdminCP? You can change some colors and fonts by editing your cp.css file in your forum directory.

That doesn't edit the layout, that edits the colours and font styles.

To edit the layout of that page, I believe you should go to admin/settings.php

I do not recommend you modify this file unless you know exactly what you are doing as it can cause problems ;)
